Abstract

Nitrile의 생변환에 이용되는 nitrile hydratase의 enzyme action에 영향을 주는 기질과 생산물 및 저해물질 등의 효과를 알아보았다. 기질로서 과잉의 acrylonitrile은 효소의 활성을 저해하는데, 기질농도가 비교적 높은 0.2mol/L에서부터 저해가 일어났다. Nitrile 생변환의 생산물인 acrylamide와 propionamide 및 이와 유사한 구조를 가지는 acrylic acid와 cyanide등은 nitrile hydratase를 경쟁적으로 저해하였다. Cyanide에 의한 저해가 가장 크게 나타났으며, 이 때 competitive inhibition constant Ki는 3.8×10-3mol/L이었다.
The effect of several compounds on the enzyme action of the nitrile hydratase was studied. An excess of acrylonitrile as substrate was shown to inhibit the activity of the enzyme. This inhibition occured at a substrate concentration of 0.2mol/L. The nitrile bioconversion products (acrylamide, propionamide)and their structural analogues(acrylic acid, cyanide) were shown to inhibit the enzyme competitively. The most important inhibition found was that of cyanide(Ki=3.8×10-3mol/L), a breakdown product of some nitriles.
