Abstract

미생물의 고농도 고정화 법으로서 고정화 캡슐을 사용하여 호기성 균주의 일종인 C. glutamicum을 캡슐내부에 고정화하고 배양특성을 살펴보았다. Surfactant가 포함된 calcium alginate 캡슐내에 C.glutamicum을 고정화하여 성장배지에서 배양하는 경우 배지용액 liter당 50g의 CaCo3와 10g의 CaCl2를 첨가함으로써 캡슐막의 용해를 방지할 수 있었다. 배지 내의 yeast extract 농도를 10g/l이상으로 유지함으로써 캡슐내부 건조중량을 220g/l이상으로 유지할 수 있었다. 캡슐내부에 산소전달 속도를 증가시키기 위해서 homoglobin을 미생물과 함께 고정화하였으며 이 때의 L-lysine 생산성은 약 30% 증가하고 캡슐내부의 건조중량은 약 40% 감소를 하였다. 이 효과는 고정화 캡슐을 5회 반복 사용할 때까지 유지되었다. 고정화 캡슐을 이용하여 플라스크 배양대신 산소공급 반응기 내에서 배양한 경우 150-200%의 L-lysine 생산농도 증가를 얻을 수 있었다.
Fermentation characteristics of encapsulated Corynebacterium glutamicum using calcium alginate membrane were investigated. The dissolution of calcium alginate capsule in the growth medium. C. glutamicum encapsulated at the inside of the capsule sneaked out through the pore of capsule wall membrane. However the dried cell density in the capsule was maintained as 220g/l by increasing the concentration of yeast extract to 10g/l in the growth medium. We added a little amount of hemoglobin to the capsule core to increase the oxygen transfer rate in the capsule. The production rate increased by 30% and the dried cell density in the capsule decreased by 40%. The effect of hemoglobin was maintained for five batch runs. The concentration of L-lysine increased to about 150-200% by pumping 420ml of air per minute in the reactor.
